  • Studio della fisiopatologia del diabete di tipo 2, con particolare riferimento all’insulino-resistenza e al suo link con l’infiammazione, e identificazione di nuovi target terapeutici per strategie farmacologiche innovative.
  • Studio degli effetti pleiotropici dei farmaci antidiabetici (inibitori della DPP4 e di SGLT-2), in particolare saranno indagati i potenziali effetti renali e antinfiammatori

  • Studio del ruolo del recettore orfano GPR21 nella patogenesi del diabete di tipo 2 e della sua inibizione come nuova potenziale strategia farmacologica per contrastare l’insulino resistenza. 

    Abbiamo già dimostrato in cellule HepG2 che l’attività di GPR21 favorisce lo sviluppo di insulino resistenza e che la sua inibizione farmacologica tramite un agonista inverso migliora l’insulino sensibilità e induce un aumento dell’uptake del glucosio aumentando la traslocazione in membrana del GLUT-2.

Inoltre, abbiamo anche dimostrato che l’inibizione di GPR21 modula il comportamento dei macrofagi, contrastando l’attività proinfiammatoria del fenotipo M1 e stimolando la migrazione degli M2.

I prossimi passi saranno (i) valutare se l’inibizione di GPR21 è in grado di contrastare l’insulino resistenza già instaurata (in modelli cellulari) e (ii) identificare un nuovo agonista inverso più potente dell’unico al momento disponibile, GRA2.

  • Studio degli effetti renali dei DPP4 inibitori, in particolare del linagliptin, in cellule primarie epiteliali del tubulo prossimale ottenute da donatore sano (RPTEC) o paziente diabetico (DRPTEC). Sarà valutato l’effetto delle gliptine e di diversi stimoli fisiopatologici sull’attività della DPP4 e sul rilascio della sua forma solubile, sDPP4. Inoltre, sarà indagato l’effetto degli inibitori della DPP4 sul comportamento dei macrofagi.

 

  • Studio delle potenziali proprietà antinfiammatorie degli integratori alimentari. Nell’ambito di una convezione con la ditta ACFP srl, saranno investigate le proprietà anti-infiammatorie di alcuni prodotti in modelli di infiammazione indotta con LPS, andando a valutare la loro abilità nell’ inibire l’induzione di enzimi e fattori di trascrizione pro-infiammatori e il rilascio di citochine tipicamente infiammatorie. Sarà inoltre valutata la capacità di queste sostanze nel modulare l’attività di cellule tipicamente coinvolte nel fenomeno infiammatorio.

  • Study of the pathophysiology of type 2 diabetes and identification of innovative pharmacological strategies aimed to counteract insulin resistance development and to modulate impaired inflammatory pathways involved in the pathophysiology of metabolic disorders.
  • Study of pleiotropic effects of antidiabetic drugs (especially DPP-4 and SGLT2 inhibitors) by focusing on potential nephroprotective and anti-inflammatory properties.

  • Study of the involvement of the orphan receptor GPR21 in the pathogenesis of type 2 diabetes and evaluation of GPR21 inhibition as a novel pharmacological strategy to counteract insulin resistance development.
     
We have already demonstrated that GPR21 is involved in the pathogenesis of insulin resistance and that its pharmacological inhibition with an inverse agonist is able to counteract its deleterious effects by improving insulin signaling and glucose uptake in hepatic cells.

We have also demonstrated that GPR21 inhibition is able to modulate macrophages migration by inhibiting the proinflammatory activity of M1 macrophages and improving M2 migration.

Further steps will be (i) investigate if GPR21 inhibition is able to counteract the insulin resistance already established (in cellular model) and (ii) identify an inverse agonist more potent that the only one currently avalilable, GRA2.

  • Study of the potential nephroprotective effects of DPP4 inhibitors, in particular linagliptin, in primary cells of epitelial proximal tubulus derived from an healthy (RPTEC) and a diabetic donor (DRPTEC). We will evaluate the ability of gliptins and several pathofisiological stimuli to modulate the DPP4 activity and its release. In addition, the effects of DPP4 inhibitors on macrophages behaviour will be also evaluated.

  • Study of the potential antinflammatory proprieties of dietary supplements. The potential antinflammatory activity will be investigated in a model of inflammation induced by LPS, and the expression of proinflammatory enzymes, the activity of proinflammatory trascriptional factors and the citokine release will be measured in presence or absence of the dietary supplement.
