State of the art:

Cardiovascular disorders associated with metabolic diseases are referred to as cardiometabolic diseases (CMD). CMD describe a spectrum of interconnected pathological alterations in organs involved in metabolic and cardiovascular functions. CMD are multifactorial diseases with important health and economic implications. Despite the recent publication of several documents suggesting clinical and social interventions to prevent CMD, the identification of common mechanisms of disease are far from clear. Most recent evidences suggest a substantial role of a low-grade, chronic inflammatory response, known as metaflammation, in promoting CMD. Despite selective inflammatory pathways have been demonstrated to be involved in metaflammation, including the well-knonw NF-kB pathways and more innovative cascades such as NLRP3 inflammasome, so far we do not know which factor has the greatest impact and the consequences of the pharmacological modulation.

 

Aims of the project:

The general aim of the project is to identify the key inflammatory pathways involved in the cross-talk mechanisms linking metabolic diseases to cardiovascular disorders and to investigate the effects of their pharmacological modulation.

The specific objectives are:

-to identify markers of metaflammation which can be used to describe common mechanisms of CMD co-morbidities;

-to mechanistically study the role of selective inflammatory pathways in evoking metaflammation;

-to validate the proposed inflammatory pathway as pharmacological targets in CMD by testing selective pharmacological tools.
