A seldom cited shared trademark for als and Alzzeimer’s Disease (AD) is the over-expression of Calcyclin in astrocytes. Calcyclin is a 10.5 kDa Ca2+ binding protein associated with modulation of heat shock proteins, and degradation of β-catenin. A new genetic link between calcyclin and neuropathologies stems from the observation that aggregates formed in spinal cord tissue by a D40G mutation in Annexin A11 (ANXA11) are abolished by over-expression of calcyclin. The annexin gene family comprises Ca-binding proteins made up of four homologous domains, with a highly conserved C-terminus and a highly variable N-terminus. Annexin domains interact among themselves in a specific and calcium-regulated fashion, whereas the determinants of ANXA11 interactions with calcyclin are still unknown
The project aims at investigating the structural determinants of interactions within ANXA11 domains and among ANXA11 and calcyclin, and to assess their dependence on metals, including some that show altered levels in ALS patients
